Mice hemizygous for this GAD67-GFP transgene are viable and fertile. GAD67-GFP (line G42) mice selectively express enhanced green fluorescent protein (EGFP) in the calcium-binding protein parvalbumin (Pv)-expressing subclass of basket interneurons (soma, dendrites, and axons) and also in putative presynaptic boutons. While transgene expression (EGFP expression) is published as early as postnatal day 0 (P0) with high expression levels throughout postnatal development, the donating investigator additionally reports transgene expression as early as embryonic day 14 (E14). EGFP expression is not reported in other interneuron classes positive for somatostatin (SOM), cholecystokinin (CCK), calretinin (CR), and VIP. These GAD67-GFP (line G42) mice may be useful for fluorescent labeling of the Pv-expressing subset of GABAergic neurons, allowing reliable and efficient characterization of perisomatic innervation in vivo.Of note, this GAD67-GFP strain is one of many fluorescent GABAergic neuron strains, each with unique labeling characteristics (see Stock No. 003718, Stock No. 006334, Stock No. 006340, Stock No. 007614, and Stock No. 007673).
Strain Development
The bacterial artificial chromosome (BAC) clone 56I19 containing the mouse GAD67 gene (and 60 kb of upstream and downstream sequence) was modified by insertion of an Enhanced Green Fluorescent Protein (EGFP) cDNA and phosphoglycerate kinase polyadenylation sequence in the first coding exon at the translation initiation site of the GAD67 gene. This GAD67-GFP BAC was microinjected into fertilized C57BL/6 mouse eggs. Transgenic offspring with appropriate GFP expression levels and patterns (founder line G42) were established on the C57BL/6 genetic background. After many generations of breeding to C57BL/6, an increase in hydrocephalus incidence was observed on the coisogenic background. To eliminate the undesired phenotype, this colony was subsequently maintained by breeding hemizygous mice to CB6F1/J (Stock No. 100007) for multiple generations prior to arrival at The Jackson Laboratory.

| Breeding & Husbandry | When maintaining a live colony, hemizygous mice are bred to wildtype siblings or to CB6F1/J (Stock No. 100007) mice. The donating investigator reports that an increased incidence of hydrocephalus was observed when maintained as coisogneic on a C57BL/6 background. |
