Description
Homozygous females and hemizygous males carrying this X-linked K14-rtTA transgene (K14-rtTetA(X) or rtTAX) are viable and fertile. These mice express the reverse tetracycline-controlled transactivator (rtTA) protein under the control of the human keratin 14 promoter. As the transgene is located on the X chromosome, random inactivation of the X chromosome may result in mosaic rtTA expression patterns in female mice. When mated to a second transgenic strain carrying a gene of interest under the regulatory control of a tetracycline-responsive promoter element (TRE or tetO), expression of the target gene in the basal cells of the squamous epithelia and in the outer root sheath of the hair follicles is induced with administration of the tetracycline analog, doxycycline (dox). These K14-rtTA mice provide a Tet-On tool that allows the inducible expression of genes in skin cells.

Development
The K14-rtTA transgene contains the 2.3 kb fragment of the human keratin 14 promoter that drives expression of the reverse tetracycline transactivator gene (rtTA), followed by an SV40 polyadenylation sequence. The 4.9 kb transgene was injected into one-cell (C57BL/6 x SJL) mouse zygotes. Mice from founder line 208 found to harbor the transgene on the X chromosome (also called founder line K14-X) were maintained on this mixed C57BL/6;SJL background prior to arrival at The Jackson Laboratory.

Gene & Allele Information

Allele Symbol Tg(KRT14-rtTA)208Jek
Allele Name transgene insertion 208, Jeffrey E Kudlow
Allele Type Transgenic (random, expressed)
Common Name(s) K14-X; K14-rtTA; K14-rtTA(X); X-linked K14-rtTA; rtTA(X);
Mutation Made By Jeffrey Kudlow,   University of Alabama at Birmingham
Strain of Origin(C57BL/6 x SJL)
Site of ExpressionExpresses rtTA in skin.
Expressed Gene rtTA, reverse tetracycline-controlled transactivator, E. coli
The tetracycline repressor gene (Tetr), arose from chemically mutated Escherichia coli genome which was screened for tetracycline dependence (Gossen and Bujard, 1992). One mutant with a four amino acid residue change (rTetR) exhibited dependence on tetracycline for induction of the targeted gene and was used in the rtTA construct (Gossen et al, 1995). rTetr was fused at the C-terminus with the viral co-activator, virion protein 16 of the herpes simplex virus (VP-16).
Promoter KRT14, keratin 14, human
Molecular Note The K14-rtTA transgene contains the 2.3 kb fragment of the human keratin 14 promoter that drives expression of the reverse tetracycline transactivator gene (rtTA), followed by an SV40 polyadenylation sequence. The 4.9 kb transgene was injected into one-cell (C57BL/6 x SJL) mouse zygotes. Mice from founder line 208 found to harbor the transgene on the X chromosome (also called founder line K14-X). Random X inactivation may result in mosaic rtTA expression patterns in female mice. The transgene when combined with mice carrying a gene under the control of a tetracycline-responsive promoter allows doxycycline-inducible gene expression in skin cells. Standard SupplyRepository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of approximately nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within two business days following order placement.

The Jackson Laboratory's Genotype Promise

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
