A targeting construct with the SA-attB-PGKneo 4xpA-attP-lacZ-pA cassette, in which the STOP fragment is flanked by attB and attP sites, was inserted into the Gt(ROSA)26Sor locus via electroporation of 129S4/SvJaeSor-derived AK7 embryonic stem (ES) cells. Correctly targeted ES cells were identified and used to generate mutant mice. The mice were crossed to 129S4/SvJaeSor mice for 2 generations prior to their arrival at The Jackson Laboratory and then intercrossed to generate homozygotes.
