These mutant mice express the mouse codon-optimized site-specific recombinase PhiC31o, from the endogenous Gt(ROSA)26Sor locus. PhiC31o mediates irreversible DNA recombination at specific nonidentical pairs of sequence sites, attB and attP. When crossed with a strain containing a sequence of interest flanked by attB and attP sites, PhiC31o-mediated recombination results in tissue-specific deletion of the targeted sequence. Recombination efficiency is similar to that of Cre recombinase. This strain represents an effective tool for generating tissue specific-targeted mutants that would be useful in genetic engineering.In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
Development
A targeting construct with the SA-PhiC310pA-PGKneopA cassette, which contains the mouse codon-optimized site-specific recombinase PhiC31o sequence (with a C-terminal SV40 nuclear localization signal) driven by the phosphoglycerate kinase 1 (PGK) promoter, and bovine growth hormone polyadenylation (bpA) sequence was inserted into the Gt(ROSA)26Sor locus via electroporation of 129S4/SvJaeSor-derived AK7.1 embryonic stem (ES) cells. Correctly targeted ES cells were identified and used to generate mutant mice. Homozygous mice were maintained on a 129S4/SvJaeSor background prior to arrival at The Jackson Laboratory (as Stock No. 007670). Upon arrival, some mice were backcrossed to C57BL/6J (STOCK No. 664) for at least five generations to generate this congenic strain (Stock No. 007743).
