Mice hemizygous for this ROSA26-EGFP BAC transgene are viable and fertile, with widespread EGFP expression in all organs and tissues analyzed. In addition, stable and uniform EGFP expression is observed in differentiated progeny of transplanted hematopoietic stem cells (HSC) isolated from ROSA26-EGFP mice. These ROSA26-EGFP BAC transgenic mice may be useful for fluorescent labeling of cells and tissues both in vivo and in vitro, are suitable for use in histological preparations, and are appropriate as a source of fluorescently labeled donor cell populations (such as tracking hematopoietic cell fate in transplant recipients).In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
Development
The bacterial artificial chromosome (BAC) clone RP23-401D9 containing the mouse Gt(ROSA)26Sor gene (and 153 kb of upstream and 15 kb of downstream sequence) was modified by inserting a red-shifted variant Enhanced Green Fluorescent Protein (EGFP; Clontech No. 6029-1) cDNA and polyadenylation sequence at nucleotide 521 of the first exon of transcript 1. This ROSA26-EGFP BAC was microinjected into pronuclei of B6xB6D2F1 fertilized oocytes. Founders with a single copy of the transgene (founder line I1) were established and maintained by breeding with CD1 mice for at least 7 generations prior to arrival at The Jackson Laboratory (as Stock No. 007896). Upon arrival, some mice were additionally backcrossed to C57BL/6J to generate this congenic strain (Stock No. 007897).

| Molecular Note | The bacterial artificial chromosome (BAC) clone RP23-401D9 containing the mouse Gt(ROSA)26Sor gene (and 153 kb of upstream and 15 kb of downstream sequence) was modified by inserting a red-shifted variant Enhanced Green Fluorescent Protein (EGFP; Clontech No. 6029-1) cDNA and polyadenylation sequence at nucleotide 521 of the first exon of transcript 1. Founders with a single copy of the transgene (founder line I1) were maintained on a CD-1 background. EGFP is ubiquitously and reproducibly expressed in all tissues and organs examined.
