The 5 strains that comprise this NOD congenic set (Stock No.'s 007930, 007931, 007932, 007933, 007934) contain the Idd 3 region from non-NOD strains. All of these strains are viable and fertile. Stocks containing the Idd 3 region from SWR/J, A/J, or CAST/EiJ are diabetes susceptible, while the congenic stocks containing the Idd 3 region derived from C57BL/6 or CZECHI/EiJ are diabetes resistant and produce more IL2 mRNA than the diabetes susceptible strains. These strains have been used to positionally clone Idd3 on Chr. 3 and continue to be valued to study the biological effects of variation of genes within the Idd3 region and for studying long-range chromosome remodeling.

Description
This NOD congenic strain, commonly referred to as NOD.CAST-Idd3, is carrying Chr 3 alleles, SNPs rs3022948 (7.43Mb) through rs13477164, (66.638Mb) derived from strain CAST/EiJ including the insulin dependent diabetes susceptibility 3 loci (Idd3). Diabetes onset and incidence among mice of this strain is reported to be similar to NOD/MrkTac. Thymocytes and splenocytes of this T1D susceptible stock produce 2 fold less Il2 mRNA than T1D protected strains (Stock No's. 007933 and 007934). This congenic strain, one of a set of 5 strains (Stock No's. 007930, 007931, 007933, 007934), has been useful to positionally clone Idd3 and to study the biological effects of variation of genes within the Idd3 region, specifically the Il2 gene and continue to be valued for studying long-range chromosome remodeling.

Development
A CAST/EiJ (Stock No.000928) congenic segment is 29.8Mb in length and is defined within SNPs rs3022948 (7.43Mb) and rs3140619 (37.3Mb) that includes the Idd3 locus on Chr 3. Idd3 contains major candidate gene IL2 as well as IL21, Tenr, Cetn4 and Fgf2. The CAST/EiJ congenic segment was introgressed onto the NOD/MrkTac background. This strain was produced by mating mice from NOD/MrkTac to CAST/EiJ followed by 5 additional crosses to NOD/MrkTac with selection for Idd3 markers. Siblings carrying the CAST/EiJ derived segment denoted by makers D3Mit329 through D3Mit274 were intercrossed to make homozygous. 5K SNP Chip Data, based on NCBI, build 36, identifies the congenic interval between SNPs rs6168642 (18.46Mb) and rs13477164 (66.15Mb) in addition a contaminating 10.9Mb CAST/EiJ region from Chr 9 defined between SNPs rs13480413 (109.73Mb) and rs6244221 (120.69MB) was identified. In 2007, the T1DR received this strain at generation N6F11.

Standard SupplyRepository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. “Each recipient institution, including its employees and other researchers under its control (RECIPIENT), of mice or services using mice from The Jackson Laboratory (TJL) agrees that such mice, descendants of those mice derived by inbreeding or crossbreeding, including unmodified derivatives of those mice or their descendants (“MICE”) shall not be: (i) used for any purpose other than the internal research of the RECIPIENT, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services with respect to MICE. Acceptance of MICE from TJL shall be deemed agreement by RECIPIENT to these conditions, and departure from these conditions requires The Jackson Laboratory’s prior written authorization.”
