Type JAX® GEMM® Strain - Congenic Additional information on JAX® GEMM® Strains. Type JAX® GEMM® Strain - Mutant Strain Type JAX® GEMM® Strain - Transgenic Mating System +/+ sibling x Hemizygote (Female x Male) Species laboratory mouse Donating Investigator Boris Reizis, Columbia University Medical Center Generation N13+ (03-MAR-08) Strain Description
Mice hemizygous for the Cd11c-cre transgene are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express Cre recombinase under the control of the mouse integrin alpha X (Cd11c) promoter. Cre-mediated recombination is detected in more than 95% of conventional CD11chigh dendritic cells both from lymphoid organs and from non-lymphoid tissues such as lung and epidermis, and in 50-80% of plasmacytoid dendritic cells. The dendritic cell compartment of transgenic mice is normal. Relatively low amounts of recombination are detected in lymphocytes (<10%), NK cells (12%), and myeloid cells (<1%). No increase of recombination frequency was observed in CD11clow- activated T cells. These Cd11c-cre transgenic mice (as well as CD11c-Cre-GFP transgenic mice (see Stock No. 007567)) are an effective tool for generating tissue-specific targeted mutants for studying dendritic cell homeostasis and function.Strain Development
The expression cassette encoding Cre recombinase and a polyA signal sequence was introduced into the first exon of the mouse integrin alpha X (Cd11c) gene (Itgax), within the bacterial artificial chromosome (BAC) clone RP24-361C4. The resulting ~160 Kb transgenic construct was introduced into B6CBAF1/J donor eggs. Founder line 1-1 was consequently established. The mice were crossed to 129S6/SvEvTac for several generations and then backcrossed to C57BL/6 for more than 12 generations prior to arriving at The Jackson Laboratory

| Site of Expression | most conventional CD11chigh dendritic cells and many plasmacytoid dendritic cells; low expression in lymphocytes, NK cells, and myeloid cells | ||
| Expressed Gene | cre, cre recombinase, bacteriophage P1 | ||
| Cre recombinase is an enzyme derived from the bacteriophage P1 that specifically recognizes loxP sites. Cre has been shown to effectively mediate the excision of DNA located between loxP sites. After the excision event, the DNA ends recombine leaving a single loxP site in place of the intervening sequence. | |||
| Promoter | Itgax, integrin alpha X, mouse, laboratory | ||
| Molecular Note | A transgenic construct containing sequence encoding Cre recombinase under the control of the mouse integrin alpha X (Cd11c) promoter, and a bovine growth hormone poly A signal sequence, was introduced into B6CBAF1/J donor eggs. Founder line 1-1 was consequently established. Cre recombinase expression is detected in CD8-, CD8+ dendritic cells, tissue derived dendritic cells from lymph nodes, lung and epidermis and plasmacytoid dendritic cells. Background recombination is detected in lymphocytes (<10%) and myeloid cells (<1%). [MGI Ref ID J:125869] | ||

| Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within 48 hours of order placement. |
