Strain Description
Mice homozygous for the Cd74 targeted mutation are viable and fertile. Cd74 deficiency affects CD4 T cell development; therefore these mice have very few CD4+ T cells. Antigen presenting cells have reduced, though not abolished, ability to present MHC class II antigens, such as GAD. These mice are protected from diabetes. This stock is important for investigation of MHC class II molecule associated self-antigen selection and the importance of self-peptide in CD4 T cell selection and development.

Strain Development
Cd74, antigen (invariant polypeptide of major histocompatibility complex, class II antigen-associated), located on Chromosome 18, 32cM is commonly known as Ii (Ia-associated invariant chain). Cd74 acts as an essential chaperone to promote MHC class II surface expression, Ag presentation, and selection of CD4+ T cells. A neomycin resistant cassette replaced exon 2 and 3 of the Cd74 gene. The B6.129S2-Cd74Tm1Doi was crossed to NOD/Caj for 12 generations prior to making homozygous for the mutation. In 2008, the T1DR received this strain at N12F20.

Standard SupplyRepository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand.
