These mice express a tau (MAPT)-sapphire green fluorescent protein (GFP) under the transcriptional control of the mouse melanocortin 4 receptor (Mc4r) promoter. Central nervous system distribution of GFP-producing cells is identical to that of Mc4r mRNA in wild-type mice and nearly all GFP-producing cells coexpress melanocortin 4 mRNA. This strain may be useful in studying the role of melanocortin signaling in the regulation of feeding behavior and autonomic homeostasis. Hemizygotes are viable and fertile and do not display any gross physical or behavioral abnormalities.Development
A transgenic BAC vector carrying Tau (MAPT)-Sapphire GFP in the ATG site of a mouse melanocortin 4 receptor (Mc4r) gene was introduced to B6CBAF1 embryos. Line 21 was backcrossed ten times to C57BL/6 by the donating laboratory.

| Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. In some constructs the original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source. | |||

| Molecular Note | A bovine gene encoding Tau protein was fused to a gene encoding a blue-shifted variant of GFP known as Sapphire. This gene was put under the control of 50kb of Mc4r promoter sequence and 95kb of downstream sequence. Two lines were characterized with line 21 having five insertion events and line 30 having one. Reporter gene expression and endogenous Mc4r gene expression co-localized in cells of the brain as determined by in situ hybridization.
