Description
These transgenic mice express the reverse tetracycline-controlled transactivator (rtTA) protein from the forkhead box A2 (Foxa2) locus. The expression pattern mimics endogenous (Foxa2) expression. Expression is detected in definitive endoderm, notochord and floorplate of the embryo. When mated to a second transgenic strain carrying a gene of interest under the regulatory control of a tetracycline-responsive promoter element (TRE), expression of the target gene may be regulated by the tetracycline analog, doxycycline (dox); in the presence of dox, transcription of the target gene is induced in cells where rtTA is expressed. Mice that are homozygous for this transgenic insert are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. This strain provides a tool that allows the inducible expression of genes in cells expressing FOXA2 for cardiac and neural development research.

Development
A targeting vector containing an FRT-site flanked neo selection cassette downstream of the reverse tetracycline regulatable transactivator protein (rtTA) gene and an IRES was inserted into the 3' UTR. The construct was electroporated into 129-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to Flp recombinase expressing mice of an unknown genetic background to remove the selection cassette. The mice were then intercrossed to generate homozygotes. The Flp recombinase allele was bred out of the strain.

The tetracycline-resistance gene (TetR), arose from chemically mutated Escherichia coli genome which was screened for tetracycline dependence (Gossen and Bujard, 1992). TetR was fused at the C-terminus with the viral co-activator, virion protein 16 of the herpes simplex virus (VP-16). The tetracycline-inhibitable transcription factor is a component of a bigenic system that allows doxycycline (a tetracycline analog) regulatable expression of genes that are under the direction of the tetracycline responsive promoter (TetOp)promoter.
Molecular Note The high affinity variant of the reverse tetracycline controlled transactivator, together with an IRES, was inserted into the 3' UTR of the locus. The frt-flanked neo that was included in the vector downstream of the rtTa was removed by Flp recombinase expression. [MGI Ref ID J:119721]

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control.
