These targeted mutation mice carry a tamoxifen-inducible Cre cassette knocked into the 3' UTR of the gene. When crossed with a strain containing a loxP site flanked sequence of interest, the offspring are useful for generating 4-hydroxytamoxifen-induced, Cre-mediated targeted deletions specifically in cholinergic neurons. Heterozygotes and homozygotes are normal in size, viability and fertility.Development
An IRES-creER cassette followed by an Frt-PGK-Neo-Frt selectable marker was knocked in to the 3' UTR of the gene using a 129-derived JHMI-ES1 embryonic stem cell line. The PGK-Neo was removed by crossing to a C57BL/6 background Flp recombinase strain. Flp was then bred out of the line. This strain was maintained on a mixed C57BL/6 and 129 background by the donating laboratory.

When crossed with a Tg(CAG-Bgeo/ALPP)1Lbe reporter strain and given tamoxifen, cre expression was detected in amacrine cells of the retina, and in a sparse number of cholinergic neurons in the brain.
