These transgenic mice express the mouse apolipoprotein E gene, Apoe, under the control of the mouse Cyp21a1, cytochrome P450, family 21, subfamily a, polypeptide 1, promoter. Transgene expression is specific to the adrenal gland. When crossed to B6.129P2-Apoetm1Unc/J (Stock No. 002052), transgene mRNA is not detected in the liver, testes or peritoneal macrophages. Founder line 619 mice, hemizygous for the transgene and homozygous for the Apoetm1Unc allele, express 1.6 pg Apoe mRNA per mg total RNA in the adrenal gland. This transgenic strain carries only the transgene. This mutant mouse strain may be useful in studies of atherosclerosis and hypercholesterolemia.Development
A transgenic construct containing the mouse apolipoprotein E gene under the control of the mouse Cyp21a1, cytochrome P450, family 21, subfamily a, polypeptide 1, promoter was injected into fertilized FVB/N mouse eggs. Founder line 619 animals were bred to homozygous B6;129-Apoe tm1Unc mice for 2 generations. The double mutant mice were then crossed to B6.129P2-Apoe tm1Unc/J (Stock No. 002052) for nine generations. This transgenic strain was generated by selectively breeding for the transgene from the double mutant strain.

| Molecular Note | A construct containing approximately 6 kb of the mouse Cyp21a1 promoter attached to the mouse Apoe gene was used for injection into fertilized FVB/N oocytes. Ten independent founder lines were generated. Line 619 was donated to the Jackson Laboratory andis representative of low expressing Apoe lines (lines 619 and 620). Only adrenal expression of Apoe was detected for these lines with Line 619 having the lowest mRNA expression (1.6 pg Apoe mRNA per microgram of total RNA).
