These mice contain an Enhanced Green Fluorescent Protein (EGFP) gene inserted in-frame into the Fmn1 locus. Expression of the EGFP is detected in the cytoplasm and localized along microtubules. This strain serves as a reporter strain, with EGFP expression mimicking endogenous expression patterns. Mice that are homozygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. This mutant mouse strain may be useful in studies of cytoskeleton regulation, cell motility, and vesicle endocytosis.Development
A targeting vector containing a loxP site flanked PGKneo cassette and sequence encoding EGFP (Green Fluorescent Protein) was inserted upstream of exon 6. The endogenous Fmn1 promoter drives expression of the EGFP through the in-frame insertion of the EGFP coding sequence to exon 6 of the Fmn1 gene, generating a fusion gene product (protein). The construct was electroporated into 129S6/SvEvTac derived TC-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to Cre recombinase expressing mice to remove the PGKneo cassette. The mice were crossed to C57BL/6J, and then backcrossed to 129 for 10 generations before arriving at The Jackson Laboratory. Upon arrival, mice were bred to 129S1/SvImJ inbred mice (Stock No. 002448) for at least one generation to establish the colony.
