| These transgenic mice express a myc peptide tagged mutant (truncated) human DISC1 gene under the control of tetracycline operator (tetO). When bred with another transgenic mouse expressing reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA) to create a bitransgenic animal, tissue-specific transgene expression can be regulated with the tetracycline analog, doxycycline. When bred to a strain expressing rtTA or tTA in brain tissues (see Stock No. 003010, B6;CBA-Tg(Camk2a-tTA)1Mmay/J, for example), this mutant mouse strain may be useful in studies of psychiatric disorders such as schizophrenia. | |||||||||||||||||||

These transgenic mice express a myc peptide-tagged, mutant (truncated) human DISC1 (disrupted in schizophrenia 1) gene under the control of tetracycline operator (tetO; also called tetracycline-responsive element (TRE) or tet-operator). The Donating Investigator reports that mice homozygous for the targeted mutation are viable, normal in size and do not display any gross physical or behavioral abnormalities. When bred with another transgenic mouse expressing reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA) to create a bitransgenic animal, tissue-specific mutant DISC1 transgene expression can be regulated with the tetracycline analog, doxycycline. When bred to a strain expressing rtTA or tTA in brain tissues (see Stock No. 003010, B6;CBA-Tg(Camk2a-tTA)1Mmay/J, for example), this mutant mouse strain may be useful in studies of psychiatric disorders such as schizophrenia.Development
The Tg(TRE-CMVhDISC1) transgene was designed with the myc peptide tagged mutant human DISC1 (disrupted in schizophrenia 1) gene, under the control of tetracycline operator (tetO; also called tetracycline-responsive element (TRE) or tet-operator) sequence and a cytomegalovirus minimal promoter. This transgene was injected into fertilized B6;SJL mouse eggs. Founder line 1001 was subsequently established. The mice were then crossed to CBA and possibly C3H.

involves: C57BL/6 * CBA * SJL
- behavior/neurological phenotype
- abnormal social/conspecific interaction
- male mutants spend significantly less time in non-aggressive social interaction with other males (MGI Ref ID J:146881)
- increased aggression towards mice
- males display increased aggressiveness toward other males, with an increase in number of attacks (MGI Ref ID J:146881)
- abnormal spatial reference memory
- spatial memory in females is impaired in probe trials, with females spending less time in the target quadrant where the platform had previously been (MGI Ref ID J:146881)
- hyperactivity
- male mutants show higher degree of spontaneous locomotor activity than wild-type littermate controls during a 20-hour period (MGI Ref ID J:146881)
- nervous system phenotype
- abnormal lateral ventricle morphology
- volume is significantly increased, with no change in total brain volume (MGI Ref ID J:146881)
- abnormal neuron morphology
- cultured control neurons grow more complex neurite arbors than mutant neurons (for 20-100 um rings centered on neuron soma) (MGI Ref ID J:146881)
