Mating System Noncarrier x Hemizygote (Female x Male) 30-MAY-09 Species laboratory mouse Generation N17+ (29-MAY-09) Donating Investigator F.W. Van Leeuwen, University Maastricht Description
These transgenic mice express the mutant human ubiquitin B, UBB+1, under the direction of the mouse Camk2a, calcium/calmodulin-dependent protein kinase II alpha, promoter. In hemizygotes, mutant transcript is expressed at 49% of the endogenous levels. The transgene is expressed in neurons of the cortex, hippocampus, amygdala and striatum. Hemizygous transgenic mice exhibit diminished ubiquitin-proteasome system activity with a resulting increase in ubiquitinated proteins in the cortex. Transgenic mice display a reduced synaptic plasticity in the hippocampus and impaired spatial learning. Mice homozygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical abnormalities. This mutant mouse strain may be useful in studies of neurodegenerative diseases such as Alzheimer's and Huntington disease.Development
A transgenic construct containing cDNA encoding the mutant human ubiquitin B, UBB+1, under the direction of the mouse Camk2a, calcium/calmodulin-dependent protein kinase II alpha, promoter was injected into fertilized C57BL/6 mouse eggs. Founder line 3413-1 was subsequently established. The mice were backcrossed to C57BL/6 for 16 generations before arriving at The Jackson Laboratory.

Tg(Camk2a-UBB*)3413-1Fwvl/?
C57BL/6-Tg(Camk2a-UBB*)3413-1Fwvl
- behavior/neurological phenotype
- abnormal spatial reference memory (MGI Ref ID J:147938)
- 9-month old male mice exhibit poor performance in the Morris water maze task in probe trials done 1 week after training commenced; mice show no quadrant preference for the former location of the platform, in contrast to wild-type mice, and significantly larger distance to platform travel compared to wild-type
- impaired contextual conditioning behavior (MGI Ref ID J:147938)
- in a fear-conditioning paradigm, 9-month old mice exhibit significantly less freezing behavior on day 8 after training than wild-type littermates; however, cued-conditioning behavior is similar in transgenic and wild-type littermates

| Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of approximately nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within two business days following order placement. |
