Mice hemizygous for this mNF-H-cre transgene are viable and fertile, with nuclear localized-Cre recombinase expression directed to neurons, coinciding with late stages of brain development, by the mouse neurofilament-H gene (Nefh) promoter. The cre expression in neurons (but not astrocytes) of the brain and spinal cord is mosaic with highest expression in the cortex and hippocampus. Cre recombinase activity begins around embryonic day (E)16.5-E18.5, with significantly robust activity observed after one week of age. The donating investigator reports that mNFHcre mouse line 12 homozygotes develop a neurological phenotype with seizures and become infertile and moribund. When bred with mice containing a loxP-flanked sequence, Cre-mediated recombination will result in deletion of the floxed sequences in the resulting offspring.Development
The mNF-H-cre transgene was designed with the 8.5 kb promoter fragment from the mouse neurofilament-H gene (Nefh [or NF-H]; containing the 5' UTR and 40 bp of noncoding sequence of exon 1, ending just before the start codon) upstream of a Cre recombinase gene (with SV40 nuclear localization signal and polyA signal). This transgene was injected into the pronuclei of fertilized FVB/N oocytes. The resulting transgenic offspring were bred to FVB/N (NCI Frederick, MD) mice to establish founder line 12 (also called Tg mNF-H-cre 12 line mice or mNFHcre mouse line 12). Transgenic mice were then bred to floxed-Cdk5 mice (on a mixed C57BL/6;129/SvJ genetic background; see Stock No. 003536). The resulting offspring were bred together, and some of the mice harboring the mNFHcre line 12 transgene were bred together to select away from the Cdk5 mutation. Next, mNFHcre mouse line 12 (on a mixed C57BL/6;129/SvJ;FVB/N genetic background and no longer harboring the floxed-Cdk5) were bred to wildtype siblings for approximately 15 generations prior to arrival at The Jackson Laboratory. Upon arrival, mNFHcre mouse line 12 were bred with FVB/NJ inbred mice (Stock No. 001800) for at least one generation to establish the colony.

We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. The total number of animals provided, their gender and genotype will vary. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 13 and 16 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility. If the mice are lost after the first three days (post-arrival) or do not produce progeny at your facility, a new order and fee will be necessary.Cryorecovery to establish a Dedicated Supply for greater quantities of mice.
