Type Congenic; Targeted Mutation; Additional information on Genetically Engineered and Mutant Mice. Visit our online Nomenclature tutorial. Additional information on Congenic nomenclature. Species laboratory mouse Donating Investigator Rulang Jiang, University of Rochester Description
Mice homozygous for this Bambiflox allele are viable and fertile, with loxP sites flanking exon 1 of the targeted gene. When bred to mice that express Cre recombinase, the resulting offspring will have the sequences encoding the endogenous signal peptide deleted in the cre-expressing tissue(s); this is expected to produce a null allele. These mutant mice may be useful in generating conditional mutations for studying the role of Bambi in developmental biology and the TGF-beta pathway.Development
A targeting vector was designed to insert a loxP site upstream of exon 1, and a second loxP site followed by an frt-flanked PGK-neo cassette just downstream of exon 1 of the targeted gene. The construct was electroporated into 129S1/Sv-derived CJ7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts and chimeric males were bred with C57BL/6J females to establish the colony. Mice heterozygous for the Bambiflox allele were bred together to generate homozygotes prior to arrival at The Jackson Laboratory Repository. Upon arrival, mice were bred to C57BL/6J (Stock No. 000664) for at least one generation to establish the colony.
