Description
These mice contain "STOP-hPLAP", the human alkaline phosphatase, placental (Regan isozyme), ALPP, gene under the control of the CAG (chicken beta actin promoter/enhancer and cytomegalovirus immediate-early enhancer) promoter, inserted into the Gt(ROSA)26Sor locus. Expression of the ALPP gene is blocked by a loxP-flanked STOP fragment placed between the ALPP sequence and the Gt(ROSA)26Sor promoter. In the absence of Cre recombinase, no ALPP staining is detected in all tissues examined so far including the central and peripheral nervous system, skin, muscle, lung and heart. This strain serves as a reporter strain, with successful Cre-mediated excision being indicated by ALPP expression in cre-expressing tissues. Mice that are homozygous for the targeted mutation are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. This mutant mouse strain may be useful in studies of cell lineage tracing, and is particularly useful for labeling and tracing the morphology of neurons because ALPP is a GPI-anchored membrane protein that is transported and expressed on the membrane of axons and dendrites of neurons.

Development
A targeting vector containing the human ALPP alkaline phosphatase, placental (Regan isozyme) gene driven by the CAG (chicken beta actin promoter/enhancer and cytomegalovirus immediate-early enhancer) promoter, a STOP fragment, and polyadenlyation signal sequence was inserted into the Gt(ROSA)26Sor locus. The construct was electroporated into 129P2/OlaHsd derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to C57BL/6 mice, and then backcrossed to C57BL/6 for one generation before being made homozygous. Upon arrival at The Jackson Laboratory the mice were crossed with C57BL/6J once to establish the colony.

JAX® Mice, Products & Services Conditions of Use

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.

No Warranty

MICE, PRODUCTS AND SERVICES ARE PROVIDED “AS IS”. In case of dissatisfaction for a valid reason and claimed in writing by a purchaser within ninety (90) days of receipt of mice, products or services, JACKSON will, at its option, provide credit or replacement for the mice or product received or the services provided.

In no event shall JACKSON, its trustees, directors, officers, employees, and affiliates be liable for any causes of action or damages, including any direct, indirect, special, or consequential damages, arising out of the provision of MICE, PRODUCTS or services, including economic damage or injury to property and lost profits, and including any damage arising from acts or negligence on the part of JACKSON, its agents or employees.
