Generation DefinitionsDonating Investigator Dr. Mario R Capecchi, University of Utah; HHMI Description
These R26RDTA (or ROSA26-DTA176) mice carry a loxP-flanked stop cassette associated with an attentuated diptheria toxin. When crossed with a Cre recombinase-expressing strain, tissue/cell-specific ablation of cells can be achieved. Mice that are homozygous for this floxed allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities.Development
A loxP-flanked stop cassette followed by the coding sequence for an attenuated diphtheria toxin A chain (tox 176) were introduced to the Gt(ROSA)26Sor gene. The targeting construct was electroporated into (129X1/SvJ x 129S1/Sv)F1- Kitl+-derived R1-45 embryonic stem (ES) cells. This strain was maintained on a mixed C57BL/6 and 129 genetic background by the donating laboratory.
