Mice that are homozygous for the targeted mutation are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. In response to endoplasmic reticulum (ER) stress induced by tunicamycin, homozygous mice exhibit a reduced expression of the acute phase response proteins C-reactive protein, serum amyloid P-component and serum amyloid A3. This mutant mouse strain may be useful in studies of the systemic inflammatory response to ER stress.Development
A targeting vector was designed to place a loxP site between exons 11 and 12 and FRT-flanked neomycin resistance cassette with a loxP site between exons 3 and 4. The construct was electroporated into 129S1/SvImJ and 129X1/SvJ derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to CD1 mice. To remove the floxed neo cassette and exons 4 through 11, mice were crossed to B6.FVB-Tg(EIIa-cre)C5379Lmgd/J (Stock #003724). Mice retain a single loxP site 3 prime to exon 3. Upon arrival, mice were bred to C57BL/6J for at least 1 generation to establish the colony.

| Molecular Note | A loxP site and a frt-flanked neomycin resistance cassette were inserted between exons 3 and 4, and a second loxP site was inserted between exons 11 and 12. The floxed region included the neo cassette. Mice were crossed with mice carrying Tg(Ella-cre)C5379Lmgd to remove exons 4 through 11 and parts of introns 3 and 11. The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description.
