The donating investigator claims homozygous mice are viable and fertile. These Prl-tTA transgenic mice have expression of the tetracycline-controlled transactivator (tTA) protein directed to PRL-producing cells (pituitary lactotropes/mammotropes) by the rat prolactin (Prl) promoter. The donating investigator also reports some expression in testis. When mated to a mutant strain carrying a gene of interest under the regulatory control of a tetracycline-responsive promoter element (TRE, TetRE or tetO), expression of the target gene in the PRL-producing cells may be conditionally abolished with administration of the tetracycline analog doxycycline (dox) in the double mutant offspring. These Prl-tTA transgenic mice are a Tet-Off tool that allows dox-conditional expression of genes in PRL-producing cells/tissues (such as pituitary lactotropes/mammotropes) and may be useful for studying pituitary hormone secretion and proliferation.Development
The PRL-tTA transgene was designed with a 3.2 kb promoter sequence from the rat prolactin (Prl) gene, a tetracycline-regulated transactivator (tTA) gene, and an SV40 intron/polyA sequence. This 5.2 kb transgene was microinjected into one-cell (C57BL/6 x SJL) mouse zygotes. Mice from founder line 6-5 were bred with C57BL/6;SJL wild-type mice to establish the colony. The donating investigator reports that PRL-tTA transgenic mice were bred together or to tetO-transgenic mice (on the same mixed C57BL/6;SJL genetic background) for many generations. Mice harboring only the PRL-tTA transgene were then sent to The Jackson Laboratory. Upon arrival, transgenic mice were bred with B6SJLF1/J hybrid mice (Stock No. 100012) for at least one generation to establish this colony.

| The tetracycline-resistance gene (TetR), arose from chemically mutated Escherichia coli genome which was screened for tetracycline dependence (Gossen and Bujard, 1992). TetR was fused at the C-terminus with the viral co-activator, virion protein 16 of the herpes simplex virus (VP-16). The tetracycline-inhibitable transcription factor is a component of a bigenic system that allows doxycycline (a tetracycline analog) regulatable expression of genes that are under the direction of the tetracycline responsive promoter (TetOp)promoter. | |||
