Description
Mice hemizygous for this B1-EGFP transgene are viable and fertile, with expression of enhanced green fluorescent protein (EGFP) directed to epithelial cells of the kidney, epididymis, and lung by the human V-ATPase B1 subunit (ATP6V1B1) promoter region. Specifically, EGFP is expressed at high levels in renal type A and type B intercalated cells, all epididymal narrow and clear cells, and some nonciliated airway epithelial cells. While no EGFP expression is detected in collecting duct principal cells or epididymal principal cells, about half of the renal connecting tubule principal cells show EGFP expression.

Development
The B1-EGFP transgene was designed with ~7 kb sequence from the 5' flanking region of the human V-ATPase B1 subunit (ATP6V1B1) gene (extending to but excluding the endogenous translational start codon) upstream of an enhanced green fluorescent protein (EGFP) coding region, and all followed by an SV40 early region polyadenylation signal. This transgene was injected into the pronuclei of single-cell (C57BL/6xCBA)F1 embryos. The resulting transgenic offspring, founder mouse F1 and F16, were found to have at least 100 copies per cell equivalent of the transgene. Mouse F1 and F16 were bred to (C57BL/6xCBA)F1 mice to establish founder line 1 and line 3, respectively. These B1-EGFP transgenic mice were then maintained on a mixed C57BL/6;CBA genetic background by breeding transgenic mice with wildtype mice for many generations prior to sending to The Jackson Laboratory Repository. Upon arrival, mice were bred with B6CBAF1/J hybrid mice for at least one generation to establish the colony.

Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. In some constructs the original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source.

Molecular Note The B1-EGFP transgene was designed with ~7 kb sequence from the 5' flanking region of the human V-ATPase B1 subunit (ATP6V1B1) gene (extending to but excluding the endogenous translational start codon) upstream of an enhanced green fluorescent protein (EGFP) coding region, and all followed by an SV40 early region polyadenylation signal. The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.

JAX® Mice, Products & Services Conditions of Use

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.

No Warranty

MICE, PRODUCTS AND SERVICES ARE PROVIDED “AS IS”.
