When these transgenic mice are crossed with a tissue or cell-specific cre strain to remove a floxed stop cassette, tetracycline-controlled transactivator (tTA) is expressed in hippocampal CA3 and the dentate gyrus under the control of the calcium/calmodulin-dependent protein kinase II alpha (Camk2a) promoter.When further mated to a transgenic strain that carries a gene of interest driven by a tetracycline-responsive promoter element (TRE; tetO), expression of that gene can be conditionally regulated by the presence or absence of doxycycline in the drinking water. Expression in tTA/tetO animals can be reversibly inhibited by the presence of doxycycline or induced by withdrawal of the tetracycline analog.
Development
A Camk2a (αCamKII) promoter followed by a loxP-flanked STOP cassette and tetracycline-controlled transactivator (tTA) were introduced to C57BL/6 embryos. This strain was created and maintained on a C57BL/6 background (see SNP note below) by the donating laboratory.A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 27 markers throughout the genome suggested a C57BL/6 genetic background, 3 of 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6J ; C57BL/6N genetic background.

| The tetracycline-resistance gene (TetR), arose from chemically mutated Escherichia coli genome which was screened for tetracycline dependence (Gossen and Bujard, 1992). TetR was fused at the C-terminus with the viral co-activator, virion protein 16 of the herpes simplex virus (VP-16). The tetracycline-inhibitable transcription factor is a component of a bigenic system that allows doxycycline (a tetracycline analog) regulatable expression of genes that are under the direction of the tetracycline responsive promoter (TetOp)promoter. | |||

| Molecular Note | The transgene construct contained the tetracycline transactivator (tTA) coding sequence downstream of a loxP-STOP-loxP fragement.
