Former Names B6.Cg-Tg(Pbsn-Hpn)DVv/J (Changed: 23-SEP-09 ) Type Congenic; Mutant Strain; Transgenic; Additional information on Genetically Engineered and Mutant Mice. Visit our online Nomenclature tutorial. Additional information on Congenic nomenclature. Species laboratory mouse Donating Investigator Valeri Vasioukhin, Fred Hutchinson Cancer Research Center Description
Hemizygous mice are viable and express mouse hepsin (Hpn) and humanized Renilla Green Fluorescent Protein under the control of the rat probasin promoter. Tissues in the transgenic animals show GFP fluorescence patterns consistent with the hepsin transgene expression, specifically in the ventral lobe of the prostate. Immunofluorescence staining with antibodies specific to basement membrane, histological and electron microscopical examination suggest that the basement membrane in the prostate gland is severely compromised. This mutant mouse strain may be useful in studies of prostate carcinogenesis.Development
A transgenic construct containing a modified rat probasin (Pbsn) promoter, beta globin intron, mouse hepsin (Hpn) full length cDNA , and an IRES-humanized Renilla (hr)GFP cassette was injected into C57BL/6 X CBA fertilized embryos. Founder line D was subsequently established. This line has been backcrossed to C57BL/6 for at least 6 generations.

involves: C57BL/6J * CBA
- reproductive system phenotype
- abnormal prostate gland morphology (MGI Ref ID J:92562)
- 75% of transgenic mice exhibit separation between epithelial and stromal cell layers in contrast to controls
- small areas of separation is observed in 14-week old mice
- ultrastructural analysis of prostatic basement membranes shows microblisters separating the epithelial and stromal cell compartments; disruption is at the level of the basement membrane with compromised adhesion between collagen fibers

| Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. In some constructs the original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source. | |||

Please note: Estimated available for sale dates are provided to keep customers better informed on strains under development. Please note that our Colony Managers routinely monitor the target date and edit it based on breeding performance and other factors. The length of time it takes to make a new strain available for sale depends on genotype, age, number of animals sent by the Donating Investigator, breeding performance, additional strain development (backcrossing, making homozygous), and anticipated demand for the strain/interest registered.
