These mice possess loxP sites on either side of the exons encoding the first and second immunoglobulin domain of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have the exons encoding the first and second immunoglobulin domain deleted in the cre-expressing tissue(s).When bred to a strain with Cre recombinase expression in adrenal tissue (see Stock No. 006364 for example), this mutant mouse strain may be useful in studies of steroidogenesis.
Development
A FRT site flanked targeting vector containing a PGKneo cassette was utilized in the construction of this mutant. A loxP site was inserted 500 bp upstream of the exon that encodes the first immunoglobulin domain. A second loxP site was inserted 2,100 bp downstream from the exon encoding the second immunoglobulin domain. This construct was electroporated into 129S4/SvJaeSor derived AK7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric male animals were crossed to mice carrying the Gt(ROSA)26Sortm1(FLP1)Dym allele (on the 129S4/SvJaeSor genetic background) to remove the FRT-flanked selection cassette. Mice that retained the loxP site flanked exons were then bred to 129S4/SvJaeSor mice to remove the FLP1 recombinase allele. Upon arrival at The Jackson Laboratory the mice were bred to 129S4/SvJaeJ inbred mice (Stock No. 009104) at least once to establish the colony.
