These ROSA:LNL:tTA mutant mice harbor a conditional mutation of the Gt(ROSA)26Sor locus with a loxP-flanked STOP cassette preventing transcription of the downstream optimized tetracycline-controlled transactivator protein. Applying both Cre-lox and Tet-Off technologies, these mutant mice may be useful for generating compound mutant mice in which expression of a tetracycline-responsive promoter element-driven gene of interest can be both directed to the cell types defined by the chosen Cre recombinase expression, as well as turned off by the addition of tetracycline (or its analog doxycycline).

Description
Mice heterozygous for the ROSA:LNL:tTA conditional mutation of the Gt(ROSA)26Sor locus are viable and fertile, with a loxP-flanked STOP cassette preventing transcription of a downstream optimized/modified tetracycline-controlled transactivator protein ("mtTA"). Applying both Cre-lox and Tet-Off technologies, these ROSA:LNL:tTA mutant mice may be useful to generate compound mutant mice in which expression of a tetracycline-responsive promoter element (TRE; also called tet-operator or tetO)-driven gene of interest can be both directed to the cell types defined by the chosen Cre recombinase expression, as well as turned off by the addition of tetracycline (or its analog doxycycline (dox)).

Development
The ROSA:LNL:tTA targeting vector was designed to insert a loxP-flanked STOP cassette (containing a neomycin resistance cassette and transcriptional STOP element) just upstream of a optimized form of the tetracycline controlled transactivator ("mtTA") into the Gt(ROSA)26Sor locus. The donating investigator reports that the construct was electroporated into 129P2/OlaHsd-derived HM-1 embryonic stem (ES) cells, correctly targeted ES cells were injected into recipient blastocysts and the resultant chimeric mice were bred to C57BL/6J mice to establish the colony. Mutant mice on a mixed genetic background (mostly C57BL/6 with some 129 or FVB/N) were sent to The Jackson Laboratory (as Stock No. 008600). Upon arrival, some mice were bred with C57BL/6J using a marker-assisted, speed congenic approach to generate this congenic strain (Stock No. 011008).

Site of ExpressionWidespread expression of the tetracycline-controlled transactivator (tTA) protein is initiated after removal of an upstream loxP-flanked STOP cassette by Cre recombinase. Thus the expression pattern of tTA is defined by the expression pattern of Cre recombinase.
Expressed Gene tTA, tetracycline-controlled transactivator, E. coli
The tetracycline-resistance gene (TetR), arose from chemically mutated Escherichia coli genome which was screened for tetracycline dependence (Gossen and Bujard, 1992). TetR was fused at the C-terminus with the viral co-activator, virion protein 16 of the herpes simplex virus (VP-16). The tetracycline-inhibitable transcription factor is a component of a bigenic system that allows doxycycline (a tetracycline analog) regulatable expression of genes that are under the direction of the tetracycline responsive promoter (TetOp)promoter.
Molecular Note A targeting vector was designed to insert a loxP-flanked STOP cassette (containing a neomycin resistance cassette and transcriptional STOP element) just upstream of a modified/optimized form of tetracycline controlled transactivator ("mtTA") into the Gt(ROSA)26Sor locus. and chimeric mice were established. The Jackson Laboratory's Genotype Promise

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
