These BAC Lrrk2-G2019S mice express a mutant form of mouse leucine-rich repeat kinase 2 (Lrrk2-G2019S) associated with autosomal dominant, late-onset Parkinson's disease directed by the endogenous Lrrk2 promoter/enhancer regions on the BAC transgene. These mice have reduced striatal dopamine content and may be useful for studying Parkinson's disease pathogenesis and neurodegeneration elicited by the dominant toxic effects of mutant LRRK2-G2019S expression.

Description
Mice hemizygous for the BAC FLAG-Lrrk2-G2019S transgene are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These mice express a mutant form of mouse leucine-rich repeat kinase 2 (Lrrk2-G2019S) associated with autosomal dominant, late-onset Parkinson's disease directed by the endogenous Lrrk2 promoter/enhancer regions on the BAC transgene. These BAC FLAG-Lrrk2-G2019S mice "overexpress" the mouse LRRK2-G2019S mutant protein in cerebral cortex, striatum, substantia nigra, internal capsule, and hippocampus at an approximately 6-8 fold greater level than endogenous mouse Lrrk2. These mice have reduced striatal dopamine content and may be useful for studying Parkinson's disease pathogenesis and neurodegeneration elicited by the dominant toxic effects of mutant LRRK2-G2019S expression.

Development
A bacterial artificial chromosome (BAC) (#RP23-312I9) containing the entire mouse Lrrk2 (leucine-rich repeat kinase 2) gene was modified by targeted mutation of the Lrrk2 locus to harbor the Lrrk2-G2019S mutation associated with autosomal dominant, late-onset Parkinson's disease originally identified in Spanish families originating from the Basque region. This modified 240 kb BAC, containing a FLAG-tag downstream from the start codon, was microinjected into fertilized B6C3 F1 oocytes. FLAG-Lrrk2-G2019S founder line 2 was established and then subsequently maintained by breeding transgenic mice with C57BL/6J inbred mice for many generations prior to arrival at The Jackson Laboratory. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

MICE and PRODUCTS are to be used in a safe manner and in accordance with all applicable governmental rules and regulations.

The foregoing represents the General Terms and Conditions applicable to JACKSON’s MICE, PRODUCTS or services. In addition, special terms and conditions of sale of certain MICE, PRODUCTS or services may be set forth separately in JACKSON web pages, catalogs, price lists, contracts, and/or other documents, and these special terms and conditions shall also govern the sale of these MICE, PRODUCTS and services by JACKSON, and by its licensees and distributors.

Acceptance of delivery of MICE, PRODUCTS or services shall be deemed agreement to these terms and conditions. No purchase order or other document transmitted by purchaser or recipient that may modify the terms and conditions hereof, shall be in any way binding on JACKSON, and instead the terms and conditions set forth herein, including any special terms and conditions set forth separately, shall govern the sale of MICE, PRODUCTS or services by JACKSON.
