Ly6a-GFP transgenic mice have an enhanced green fluorescent protein (EGFP) under the control of murine lymphocyte antigen 6 complex, locus A (Ly6a) promoter. Hemizygous Ly6a-GFP mice are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. The Ly6a promoter directs the expression of GFP in all functional repopulating adult hematopoietic stem cells (HSCs) in the adult bone marrow, and several other hematopoietic cell types. The GFP transgene expression pattern generally corresponds to that of Sca-1, a glycoprotein-I-linked cell-surface glycoprotein used routinely as a marker of adult hematopoietic stem cells. These mice may be useful for the visualizing and sorting of hematopoietic stem cells.Development
The Ly6a-GFP transgene contains an enhanced green fluorescent protein (EGFP) and the simian virus 40 (SV40) polyadenylation (polyA) sequence under the control of murine lymphocyte antigen 6 complex, locus A (Ly6a) promoter. The construct was microinjected into fertilized (C57BL/10 x CBA)F1 oocytes, and offspring from founder line G5 were bred to (C57BL/10 x CBA)F1 mice to establish a colony. The donating investigator reported that these mice were then backcrossed to C57BL/6J (see SNP note below) for at least 10 generations. Upon arrival at The Jackson Laboratory, mice were bred with C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 27 markers throughout the genome suggested a C57BL/6 genetic background, 2 of 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6J ; C57BL/6N genetic background.
