Description
Mice hemizygous for the Lck-cre transgene are viable, fertile, and normal in size. These mice contain a codon-improved Cre recombinase gene driven by the distal promoter of the lymphocyte protein tyrosine kinase (Lck) gene. Cre recombinase expression is delayed, and is only observed in T cells after T cell receptor α (Tcra) locus rearrangement and after the cells have reached the TCRhi stage of thymocyte development. When these transgenic mice are bred with mice containing a loxP-flanked sequence, Cre-mediated recombination results in deletion of the floxed sequences in the Cre recombinase-expressing tissues of the offspring. These mice are useful for studying the consequences of mutations induced after positive selection in the thymus.

Development
The Lck-cre transgene was designed with codon-improved cre recombinase gene (icre) driven by the distal promoter of the lymphocyte protein tyrosine kinase (Lck) gene, followed by a human growth hormone cassette. This transgene was injected into (C57BL/6 x DBA) F2 fertilized oocytes. Transgenic mice from founder line 3779, with one transgene insertion site, were bred to C57BL/6J and the resulting offspring were then backcrossed to C57BL/6J for 12 generations to produce a colony of Lck-cre mice. Upon arrival at The Jackson Laboratory, these mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

JAX® Mice, Products & Services Conditions of Use

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.

MICE, PRODUCTS AND SERVICES ARE PROVIDED “AS IS”.
