These Atoh1FLAG mice contain three c-terminal FLAG tags fused to the 3' end of the atonal homolog 1 (Atoh1) gene. Homozygous mice are viable and fertile. Atoh1FLAG expression is evident evident in all Atoh1 expressing cells including cerebellar granule neuron precursors (GNPs) where it binds the active transcriptional enhancer region of the GLI-Kruppel family member GLI2 (Gli2) gene. These mice may be useful for studying postnatal cerebellar development and regulation of Sonic Hedgehog-induced medulloblastoma.Development
A targeting vector was designed to fuse three c-terminal FLAG tags to the 3' end of the atonal homolog 1 (Atoh1) gene, followed by a frt-flanked neomycin (neo) selection cassette in reverse orientation to the gene. This construct was electroporated into 129S7/SvEvBrd-Hprt1b-m2-derived AB2.2 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts and chimeric mice were bred with 129S4/SvJaeSor-Gt(ROSA)26Sortm1(FLP1)Dym mutant mice to remove the neo cassette. The resulting Atoh1FLAG mice were subsequently mated with inbred C57BL/6 mice for at least 8 generations. Upon arrival at The Jackson Laboratory, these mice were bred with C57BL/6J mice (Stock No. 000664) for at least one generation to establish the colony.

The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility.
