Description
These Prkaa1 mutant mice possess loxP sites flanking exon 3 of the protein kinase, AMP-activated, alpha 1 catalytic subunit (Prkaa1) gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Prkaa1 belongs to the serine/threonine protein kinase family and is a catalytic subunit of the 5'-prime-AMP-activated protein kinase (AMPK). AMPK is a sensor of the energy status of cells and promotes survival during stress. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues. When bred to transgenic mice expressing Cre recombinase driven by myxovirus (influenza virus) resistance 1 (Mx1) promoter/enhanced elements, PRKAA1 expression is abolished in hematopoietic cells. This strain may be useful for studying cell growth and energy expenditure.

Development
A targeting vector was designed to insert a loxP site upstream of exon 3, and a Frt-flanked neomycin resistance cassette (neo) followed by a second loxP site downstream of exon 3 of the protein kinase, AMP-activated, alpha 1 catalytic subunit (Prkaa1) gene. The construct was electroporated into Bruce4-derived C57BL/6 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts from B6(Cg)-Tyrc-2J/J mice (Stock No. 000058) and the resulting chimeric males were bred to female B6(Cg)-Tyrc-2J/J mice to establish a colony. These mice were then bred to mice carrying Tg(ACTFLPe)9205Dym to delete the neo cassette. Offspring were then backcrossed to C57BL/6 for at least 4 generations (see SNP note below). Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. At least 7 the 32 markers on 5 chromosomes were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.

MICE and PRODUCTS are to be used in a safe manner and in accordance with all applicable governmental rules and regulations.
