The mouse Per1 (period homolog 1 (Drosophila)) promoter drives in vivo and in vitro expression of luciferase in a pattern recapitulating that of circadian rhythms in this transgenic strain. These mice may be useful in studies of circadian and photic regulation of the mammalian circadian oscillator.

Description

The genotypes of the animals provided may not reflect those discussed in the strain description or the mating scheme utilized by The Jackson Laboratory prior to cryopreservation. Please inquire for possible genotypes for this specific strain.

Description
In this transgenic strain, the mouse Per1 (period homolog 1 (Drosophila)) promoter drives in vivo and in vitro expression of luciferase in a pattern recapitulating that of circadian rhythms in the suprachiasmatic nuclei (SCN). Long-term circadian rhythms of luminescence are also seen in SCN slice culture. Pulses of light rapidly induce high luciferase mRNA expression in the SCN that, unlike endogenous Per1 mRNA levels, are sustained and do not return to baseline. This strain is reported to carry 2 copies of the transgene, but copy number analysis performed at the Jackson laboratory indicates that transgenic line 128 (see Stock No. 016165) is of higher luciferase copy number than transgenic line 141 (see Stock No. 016166).

Development
6.75 kb of the mouse Per1 promoter (including exon 1, intron 1 and the untranslated portion of exon 2 up to the initiaton ATG) drives expression of luciferase in this transgenic strain. The initiator ATG of both Per1 and luciferase were engineered to coincide. The vector was introduced to CD1 embryos. This strain was backcrossed to C57BL/6J for 10 generations by the donating laboratory. This strain is reported to carry 2 copies of the transgene, but copy number analysis performed at the Jackson laboratory indicates that transgenic line 128 (see Stock No. 016165) is of higher luciferase copy number than transgenic line 141 (see Stock No. 016166).

The Jackson Laboratory's Genotype Promise

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
