In this transgenic strain, the mouse Per1 (period homolog 1 (Drosophila)) promoter drives in vivo and in vitro expression of luciferase in a pattern recapitulating that of circadian rhythms in the suprachiasmatic nuclei (SCN). Long-term circadian rhythms of luminescence are also seen in SCN slice culture. Pulses of light rapidly induce high luciferase mRNA expression in the SCN that, unlike endogenous Per1 mRNA levels, are sustained and do not return to baseline. This P1L128 line carries 2 copies of the transgene.Development
6.75 kb of the mouse Per1 promoter (including exon 1, intron 1 and the untranslated portion of exon 2 up to the initiaton ATG) drives expression of luciferase in this transgenic strain. The initiator ATG of both Per1 and luciferase were engineered to coincide. The vector was introduced to CD1 embryos. This strain was backcrossed to C57BL/6J for 10 generations by the donating laboratory. This strain carries 2 copies of the transgene.
