Description
Dnmt3l (DNA (cytosine-5-)-methyltransferase 3-like) is an enzymatically inactive regulatory factor that establishes patterns of DNA methylation inactivation during embryogenesis. A cysteine-rich domain of this protein interacts with the extreme amino terminal of histone H3. Data indicate that DNMT3L recognizes histone H3 tails that are unmethylated at lysine 4 and induces de novo DNA methylation by recruitment or activation of DNMT3A (DNA methyltransferase 3A). In this mutant, a tandem His6-Flag tag was introduced to the N terminus of the endogenous Dnmt3l mouse gene. Anti-flag immunoprecipitation can be used to isolate and analyse DNMT3L complexes (e.g. DNMT3A, DNMT3B, core histones). Unlike Dnmt3l knockout mice, both males and females carrying this knock-in allele have no overt phenotype and are viable and fertile in the homozygous state, suggesting normal functionality of the targeted gene.

Development
A targeting vector was designed to introduce a tandem His6-Flag tag immediately after the first four amino acids (encoded by exon 2) of the mouse Dnmt3l gene. A loxP-flanked PGK-neomycin cassette was placed in intron 4. The vector was introduced to 129/SvEv-derived embryonic stem cells and transient exposure to Cre recombinase excised the neomycin cassette. Resultant mice were backcrossed once to C57BL/6 to establish the colony. Mutant mice were bred together for several generations prior to sending agouti mice to The Jackson Laboratory Repository.

JAX® Mice, Products & Services Conditions of Use

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.

No Warranty

MICE, PRODUCTS AND SERVICES ARE PROVIDED “AS IS”.
