The coding region of the Nanog homeobox (Nanog) is replaced by a GFP-IRES-puro-cassette in these mice, abolishing gene expression. NANOG is normally expressed in embryonic stem cells (ESCs) and functions as a key factor in maintaining pluripotency. Heterozygous mutant mice are viable and fertile, while homozygotes are embryonic lethal. Mouse embryonic fibroblasts (MEFs) derived from these GFP reporter mice are useful for identifying induced pluripotent stem (iPS) cell formation.Development
A targeting vector was designed to replace the Nanog homeobox (Nanog) coding region with an enhanced green fluorescent protein (GFP), an internal ribosome entry site (IRES), a puromycin (puro) resistance cassette and a polyadenylation signal. This construct was electroporated into (C57BL/6 x 129S4/SvJae)F1-derived V6.5 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient (C57BL/6 x DBA)F2 blastocysts and chimeric males were bred with 129S4/SvJae females. These Nanog-GFP mice were intercrossed and maintained on a mixed background. Upon arrival at The Jackson Laboratory Repository, mice were bred with 129S1/SvImJ inbred mice (Stock No. 002448) for at least one generation.
