The dnTRPC3 transgene contains amino acids 1-302 of the human transient receptor potential cation channel, subfamily C, member 3 (TRPC3) gene under the control of the mouse myosin, heavy polypeptide 6, cardiac muscle, alpha (Myh6) promoter. Hemizygotes are viable, fertile and normal in size. TRPC3 is a nonselective monovalent cation channel which is activated by oxidative stress in endothelial cells. These mice express a dominant-negative form of TRPC3. They only express the N-terminus of the gene and lack the transmembrane and cytosolic carboxy terminal domains. TRPC3 transgenic mice exhibit an increase in cardiac calcineurin-nuclear factor of activated T cells (NFAT) transcriptional activity, cardiac hypertrophy, and cardiomyopathy. They show a dosage-dependent increase in cardiac hypertrophy following neuro-endocrine agonist or pressure overload stimulation.Development
The dnTRPC3 transgene contains amino acids 1-302 of the human transient receptor potential cation channel, subfamily C, member 3 (TRPC3) under the control of the mouse myosin, heavy polypeptide 6, cardiac muscle, alpha (Myh6) promoter. The construct was microinjected into fertilized FVB/N oocytes, and offspring from founder line 6.6 were bred to FVB/N mice to establish a colony. Upon arrival at The Jackson Laboratory, mice were bred to FVB/NJ inbred mice (Stock No. 001800) for at least one generation.

The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility.
