These mice carry a knock-out mutation of the Plk2 gene. Homozygotes exhibit a decrease in soluble alpha synuclein phosphorylation in the brain and may be useful in studies of Parkinson's Disease.

Description
Mice that are homozygous for the targeted mutation and on the 129S background are viable and fertile, the Donating Investigator reports. On the C57Bl/6NTac background, few live homozygotes are observed. No gene product (mRNA) is detected by qRT-PCR analysis of primary cortical cultures. Alpha synuclein phosphorylation is decreased by approximately 70% in cortical cultures from homozygous embryos, when compared to wildtype controls. In one 2-3 month old homozygous mouse, a similar decrease in soluble alpha synuclein phosphorylation was observed.

In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. This is the case for the strain above. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.

Development
A targeting vector containing a NEO cassette was used to disrupt all 14 exons of the Plk2, polo-like kinase 2, gene. The construct was electroporated into unspecified C57BL/6NTac derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to C57BL/6NTac mice. The mice were then backcrossed to 129S6/SvEvTac using a speed congenic protocol, for approximately 6 generations. Upon arrival at The Jackson Laboratory, the mice were crossed to 129S1/SvImJ (Stock No. 002448) at least once to establish the colony.

JAX® Mice, Products & Services Conditions of Use

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.

No Warranty

MICE, PRODUCTS AND SERVICES ARE PROVIDED “AS IS”.
