These PP2A-A-alpha-E64G mice contain the amino acid mutation E64G in exon 3 of the A-alpha subunit gene (Ppp2r1a) of protein phosphatase 2A (PP2A). This mutation was detected in a human breast carcinoma. Homozygotes are viable and fertile. PP2A holoenzyme is composed of a catalytic C subunit, a scaffolding A subunit, and a regulatory B subunit. It suppresses the development of lung cancer induced by the carcinogen benzopyrene. This tumor suppressor activity is p53-dependent. PP2A also prolongs the survival of mice expressing oncogenic K-rasG12D (v-Ki-ras2 Kirsten rat sarcoma). PP2A-A-alpha-E64G mice are a model for studying the tumor suppressor activity of B'-containing PP2A holoenzymes in mammary gland, lung, and other organs. The E64G mutation in this mouse strain renders the Aα subunit defective in binding regulatory B' subunits whereas binding of B and B'' subunits is normal. These mice may be useful for studying tumor suppression, development, and aging.Development
A targeting construct was designed to introduce two point mutations in codon 64, exon 3, of the A-alpha subunit gene (Ppp2r1a) of protein phosphatase 2A (PP2A) resulting in the amino acid substitution E64G discovered in a human breast carcinoma. A loxP-flanked neomycin (neo) resistance cassette was inserted downstream of exon 3. This targeting construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells and correctly targeted ES cells were injected into blastocysts. Chimeric males were bred to female 129S mice and the resulting offspring were subsequently bred to mice carrying prion driven Cre-recombinase on a 129S background to remove the neo cassette. These PP2A-A-alpha-E64G mice were then backcrossed for at least 14 generations to FVB/NCrl mice to establish a colony. Upon arrival at The Jackson Laboratory, these were bred to FVB/NJ (Stock No. 001800) mice for at least one generation.

The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description.
