Description
Transient receptor potential canonical (TRPC) channels are cation-selective influx channels that can initiate cardiac hypertrophy when overexpressed, in part because of increased Ca2+ influx and calcineurin activation.

Three amino acids encoding the putative pore region of the human TRPC6 gene (transient receptor potential cation channel, subfamily C, member 6 ) were replaced by alanines in this transgenic mutation driven by the mouse Myh6 (myosin, heavy polypeptide 6, cardiac muscle, alpha) promoter. Expression of this dominant negative mutation (also known as dnTRP6) suppresses transient receptor potential, thereby preventing pathological cardiac remodeling after pressure overload. This strain is useful in studies of cardiac hypertrophy.

Development
The mouse Myh6 promoter drives expression of human TRPC6 cDNA in which three highly conserved amino acids (L678-W680) within the putative pore region have been replaced by alanine (A) residues. The dominant negative dnTRP6 mutation was created and maintained on an FVB genetic background by the donating laboratory.

The Jackson Laboratory's Genotype Promise

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
