Gata4 (GATA binding protein 4) is expressed in mesoderm-derived tissues such as the heart, liver, gut and gonads. It is a critical transcription factor in the heart that is known to induce/regulate the hypertrophic program, in part, by receiving signals from MAPKs.In this targeted mutation strain, serine 105 of the mouse Gata4 gene was changed to an alanine to disrupt a known MAPK phosphorylation site. Homozygotes are viable, fertile, and born at predicted Mendelian frequencies. Mice show a compromised stress response of the myocardium. Cardiac hypertrophy in response to phenylephrine agonist infusion and pressure overload are largely blunted. Homozygotes are also more susceptible to heart failure and cardiac dilation after 2 weeks of pressure overload.
Development
Serine 105 of the targeted gene (located in exon 2) was mutated to an alanine (S105A) and an FRT-flanked neomycin selection cassette was introduced to intron 2. A 129-derived embryonic stem (ES) cell line was used to create the mutation. A cross with a germline/testes-specific FLP strain of unknown genetic background excised the neomycin cassette. This strain was backcrossed to C57BL/6 for 3 generations by the donating lab.

The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility.
