This strain of mice, first published in 1986 on a mixed C57BL/6-C3H genetic background, successfully demonstrated the transgenic expression of human insulin in the pancreas. Human pro-insulin is enzymatically cleaved into insulin and C-peptide in pancreatic islets. In these mice, no human C-peptide was detected in the sera of fasting transgenic mice, but within 10 minutes of i.p. administration of glucose, human C-peptide appeared in the serum. Peak levels were obtained within ~20 minutes. By 45 minutes post-glucose, human C-peptide levels fell to values approaching the pre-stimulation or basal level.Injected with human insulin, these mice demonstrated an acquired tolerance to human insulin and pro-insulin. These animals showed no antibody response. This model presented a novel alternative to the induction of tolerance in newborn mice using antigenic dosing.
The current line offered here has been backcrossed to BALB/cJ and is on a homozygous H2d background.
Development
A 12.5 kb EcoRI fragment containing the human insulin gene was microinjected into C57BL/6 x C3H F2 embryos by Howard Goodman's lab at Harvard University. Originally published in 1986, H2k progeny were subsequently selected by Judith Kapp's group at The Jewish Hospital of St. Louis at Washington University Medical School and backcrossed to BALB/cJ (H2d) for at least 10 generations.

The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility. If the mice are lost after the first three days (post-arrival) or do not produce progeny at your facility, a new order and fee will be necessary.Cryorecovery to establish a Dedicated Supply for greater quantities of mice.
