These Flk1-TVA transgenic mice contain the mouse kinase insert domain protein receptor (Kdr) promoter driving expression of an avian specific retroviral receptor (TVA) derived from quail. FLK1 is a receptor tyrosine kinase expressed on proliferating adult and embryonic endothelial cells, and is involved in the development of angiogenic vasculature. Hemizygous mice are viable and fertile. Expression of TVA in these mice makes them highly susceptible to infection by RCAS (replication-competent, avian leukosis virus LTR, splice acceptor) gene delivery machinery. The RCAS integrates its genetic information into the host DNA, promoting a high level of viral transcription. The insertion of a gene of interest into multicloning site within RCAS-derived constructs allows for its cell specific constitutive expression in vascular epithelial cells. These TVA mice may be useful for studying genes of interest involved in angiogenesis.Development
The Flk1-TVA transgene was designed with the kinase insert domain protein receptor (Kdr) promoter driving expression of an avian specific retroviral receptor (TVA) gene derived from quail. The transgene was microinjected into fertilized FVB/N oocytes, and mice from founder line 1026 were then bred to FVB/NJ mice to establish a colony. Upon arrival at The Jackson Laboratory, mice were bred to FVB/NJ inbred mice (Stock No. 001800) for one generation.

| Molecular Note | A 786-nucleotide cDNA (nucleotides 34-819 of pg800) encoding the quail receptor (TVA) for avian leukosis viruses of subgroup A [ALV(A)] is expressed under the control of regulatory sequences from the mouse Kdr (kinase insert domain protein receptor) gene: a -640 bp to +299 bp genomic DNA fragment containing the promoter and a 2.3 kb fragment containing the endothelium-specific enhancer from the first intron. Embryonic day (E) 10.5 embryos of this line were shown by whole mount in situ hybridization to express TVA mRNA selectively in the developing vasculature, in a pattern similar to that of endogenous Kdr mRNA. The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility.
