The mouse Serpinb3a gene is similar to the human SERPINB4 and SERPINB3 genes, which have been associated with allergic reactions and asthma. Mouse Serpinb3a inhibits both serine and cysteine proteinases. These mice carry a null targeted mutation of Serpinb3a; exon 8 is disrupted. No gene product (mRNA) is detected by RT-PCR analysis of homozygotes. Serpinb3a-deficient mice are less sensitive to house dust mite-induced airway hyper-responsiveness, with reduced goblet cell hyperplasia and mucus production when compared to wildtype controls. IL13-induced airway hyper-responsiveness is reduced in homozygotes, with a reduced number of inflammatory cells in the bronchoalveolar lavage fluid. Mice that are homozygous for the targeted mutation are viable and fertile.Development
A targeting vector containing a NEO cassette was used to disrupt exon 8. The construct was electroporated into unspecified 129 (129/SV) derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric animals were crossed to BALB/cAnNCrl mice, and then backcrossed to BALB/cAnNCrl for at least 5 generations. Upon arrival at The Jackson Laboratory, the mice were crossed to BALB/cByJ (Stock No. 001026) at least once to establish the colony.
