The t(15;17)(q22:q11) translocation results in the PML-RARA, protein promyelocytic leukemia-retinoic acid receptor alpha, fusion gene, which is a characteristic chromosomal translocation in acute promyelocytic leukemia. These mPML-PRflox mice carry a loxP-flanked PGKneo-stop cassette, PML-RARA cDNA, in the 5' UTR of the Pml, promyelocytic leukemia, gene. Mice that are homozygous for the targeted mutation are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. No Pml gene product (protein) is detected by Western blot analysis of spleen cells from homozygous animals. When bred to mice that express Cre recombinase, resulting offspring will express the fusion oncoprotein PML-PARA in the cre-expressing tissue(s).When bred to a strain with Cre recombinase expression in the myeloid cell lineage (see Stock No. 004781 for example), the resulting mice express PML/PARA mRNA in spleen cells and rare leukemia with acute promyelocytic leukemia characteristics.
When bred to a strain expressing tamoxifen-inducible Cre recombinase (see Stock No. 004682 for example), this mutant mouse strain may be useful in studies of hematopoietic stem cell renewal.
Development
A targeting vector containing a loxP-flanked PGKneo-stop cassette, PML-RARA cDNA, and 5 stop codons, was inserted into the Pml 5' untranslated region. The construct was electroporated into C57BL/6 derived B6-Blu embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The donating investigator reported that the resulting chimeric animals were crossed to C57BL/6 mice (see SNP note below). Upon arrival, heterozygous sperm was frozen. To generate the live colony, an aliquot of the frozen sperm was used to fertilize oocytes from C57BL/6J inbred females (Stock No. 000664).A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 27 markers throughout the genome suggested a C57BL/6 genetic background, all 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a C57BL/6N genetic background.
