The miR-34 family of miRNAs (miR-34a, miR-34b, and miR-34c) are expressed mainly in the testis, brain, and lung. miR-34 proteins are induced upon p53 activation and are associated with tumor suppression, aging, neurodegeneration, and spermatogenesis. In this strain, a targeting vector including Kozak sequence, lacZ and a neomycin cassette was used to disrupt Mir34a. No gene product (mRNA) is detected in MEFs (mouse embryo fibroblasts) from homozygotes by RT-PCR analysis. Homozygous Mir34a-/- mice are viable and fertile. MEFs from Mir34a-/- mice exhibit increased reprogramming efficiency and induction of iPSCs (induced pluripotent stem cell) when compared to controls. Mutant mice may have applications in studies related to somatic cell reprogramming and regulatory control of p53 pathways.Development
A targeting vector containing Kozak sequence, the lacZ gene and a FRT & loxP site flanked NEO cassette, was used to disrupt Mir34a and 20 bp flanking sequence. The construct was electroporated into C57BL/6-derived Bruce 4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to C57BL/6J mice. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
