Description
RAPGEF4 (or EPAC2) is a rap guanine nucleotide exchange factor that functions as an intracellular receptor for cyclic AMP. Epac2 mRNA is expressed predominantly in the brain and endocrine tissues. Mice that are homozygous for this genetrap/knockout allele are viable and fertile. When a Rapgef4 knockout is combined with Rapgef3tm1.1Geno (Epac1 conditional KO) (Stock No. 018389) and a global Cre recombinase-expressing strain, double homozygous knockout mice exhibit impaired long term potentiation (LTP), deficits in spatial learning, memory deficits, and abnormal social interactions. The donating investigator indicates mice tend toward obesity by two months of age. This strain may be useful in generating conditional mutants for studying cognition and social dysfunctions.

Development
A gene trap vector (Omnibank Gene Trap VICTR 48+ MTII) was inserted within intron 1 of the Rapgef4 gene and was electroporated into 129S5/SvEvBrd derived Lex-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were bred with 129S8/SvEvNimrJ mice. Male mice were sent to The Jackson Laboratory Repository in 2012. Upon arrival, males were used to cryopreserve sperm. To establish the living mouse colony, an aliquot of the frozen sperm may be used to fertilize 129S1/SvImJ oocytes (Stock No. 002448).

A 27 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, was performed on the first generation rederived living colony at The Jackson Laboratory Repository. This revealed 8 of 27 markers (one each on chromosomes 2, 3, 7, 11, 12, 13, 16 and 20) that were segregating for allele-type markers other than 129. These data suggest the mice are on a mixed genetic background of 129 and other undetermined contributions.

JAX® Mice, Products & Services Conditions of Use

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.

No Warranty

MICE, PRODUCTS AND SERVICES ARE PROVIDED “AS IS”.
