Integrins like Itgb2 (integrin beta 2) are involved with processes of cell adhesion and cell surface-mediated signaling. Mutations in this gene have been associated with leukocyte adhesion deficiency type I (LAD1).These mice express monomeric cyan fluorescent protein (mCFP) that has been knocked into/fused with the mouse Itgb2 gene. Homozygotes are viable, fertile, show no overt abnormal phenotype, and express normal levels of ITGB2. ITGB2 expression can be visualized directly by live animal imaging without antibody injection or fluorescent cell transfer. All leukocytes expressing ITGB2 express mCFP for purposes of in vivo and ex vivo imaging. These mice are a versatile tool for studying leukocyte behavior as it relates to Itbg2.
Development
mCFP was fused to the last exon (exon 16) of Itgb2 (CD18) gene along with a 6 amino acid linker (GGPVAT) and a loxP-flanked neomycin resistance cassette. The mutation was created in 129S6/SvEvTac-derived W4 embryonic stem (ES) cells. Resultant mice were crossed with a CMV-Cre mouse on a C57BL/6 background to excise the neomycin cassette. This strain was backcrossed to C57BL/6 for seven generations by the donating laboratory.
