These Tg(Csf1r-Mer-iCre-Mer)1Jwp transgenic mice express a Cre recombinase/mutant murine estrogen receptor double-fusion protein under the direction of the mouse Csf1r, colony stimulating factor 1 receptor, promoter. Tamoxifen-inducible cre activity is detected in bone-marrow-derived macrophages and yolk sac macrophages. When these Tg(Csf1r-Mer-iCre-Mer)1Jwp mice are bred with mice containing a loxP-flanked sequence of interest, tamoxifen-inducible, Cre-mediated recombination will result in deletion of the flanked sequences in Csf1r-expressing cells; making them useful for studying inflammation, metastasis and tracking myeloid lineage cells.The MerCreMer double fusion protein consists of Cre recombinase flanked on each end with a mutated murine estrogen receptor (mer) ligand binding domain (amino acids 281-599, G525R); which does not bind its natural ligand (17'-estradiol) at physiological concentrations but will bind the synthetic estrogen receptor ligands 4-hydroxytamoxifen (OHT or tamoxifen) and, with lesser sensitivity, ICI 182780. Restricted to the cytoplasm, MerCreMer can only gain access to the nuclear compartment after exposure to tamoxifen. To counteract the mixed estrogen agonist effects of tamoxifen injections, which can result in late fetal abortions in pregnant mice, progesterone may be co-administered.
Development
A transgenic construct containing tandem copies of icre/Esr1*, or Mer-icre-Mer, under the control of the mouse Csf1r, colony stimulating factor 1 receptor, promoter, was injected into fertilized FVB/N mouse eggs. Founder line 1 was subsequently established. Upon arrival at The Jackson Laboratory, the mice were crossed to FVB/NJ (Stock No. 001800) at least once to establish the colony.
