These Dckflox/flox mutant mice possess loxP sites flanking exon 3, encoding the catalytic domain, of the deoxycytidine kinase (Dck) gene. Deoxycytidine kinase is a gene involved in deoxyribonucleoside salvage metabolism for use in DNA replication. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues.For example, when bred to B6.C-Tg(CMV-cre)1Cgn/J mice (Stock No. 006054), resulting Dck deficient offspring exhibit severe defects in T cell, B cell, and erythroid development resulting from acute cell cycle arrest in early S-phase in rapidly proliferating progenitor populations.
Development
A targeting vector was designed to insert a loxP sites flanking exon 3 of the deoxycytidine kinase (Dck) gene, followed by a frt-flanked neomycin resistance (neo) cassette. The construct was electroporated into 129S4/SvJae-derived LW-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred to C57BL/6NTac mice. Offspring were bred with B6.Cg-Tg(ACTFLPe)9205Dym/J transgenic mice (Stock No. 005703) to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. These mice were bred to C57BL/6NTac mice for at least 4 more generations. Upon arrival, mice were bred to C57BL/6NJ inbred mice (Stock No. 005304) for at least one generation to establish the colony.
