Description
The progesterone receptor is a nuclear steroid receptor, ligand activated transcription factor which regulates gene expression following progesterone hormone binding. These mice contain IRES-PLAP-IRES-nuclear lacZ inserted into the 3' untranslated region (UTR) of the Pgr (progesterone receptor) locus. Reporter gene expression mimics the endogenous Pgr gene expression pattern. In progesterone receptor-expressing neurons, cell soma can be labeled by the nuclear lacZ and cell projections can be labeled by PLAP, placental alkaline phosphatase. Beta-galactosidase activity is detected in the forebrain (hypothalamic nuclei, posterodorsal medial amygdala, medial division of the posteromedial bed nucleus of the stria terminalis, various cortical areas, basal ganglia, and the dentate gyrus). Placental alkaline phosphatase activity is detected in neuronal fibers of the forebrain. Expression and function of Pgr is unaffected. Mice that are homozygous for the targeted mutation are viable and fertile.

Development
A targeting vector designed by Dr. Nirao M. Shah (University of California, San Francisco) containing IRES-PLAP-IRES-lacZ with nuclear localization signal sequence, and a self-excising neo resistance cassette (ACN) was inserted into the 3' untranslated region (UTR) of the gene. The construct was electroporated into 129/SvEv derived ES cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. The resulting chimeric mice were bred to C57BL/6J females. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control.
