Description
Ntrk2 (neurotrophic tyrosine kinase, receptor, type 2; also called TrkB) is a receptor for neurotrophin 4 (now called neurotrophin 5, Ntf5) and brain derived neurotrophic factor (Bdnf), a neurotrophin which controls aspects of mammalian nervous system development.

A tau-enhanced EGFP cassette was inserted into the first coding exon of Ntrk2 in this targeted mutant strain. Approximately 7% of adult thoracic dorsal route ganglion neurons express EGFP. These neurons have medium-diameter somata and exhibit properties Aδ-low-threshold mechanoreceptors (LTMRs): refined mechanical sensitivity, rapidly adapting responses to suprathreshold stimuli, intermediate conduction velocities, and narrow uninflected somal spikes. The neurons form longitudinal lanceolate endings associated with zigzag and awl/auchene hair follicles. Homozygotes are neonatal lethal, consistent with a prediction that this is a null allele.

Development
A tau-enhanced green fluorescent fusion protein linked to an Frt-Neomycin-Frt-loxP cassette was introduced to the first coding ATG in exon 2 by homologous recombination in 129S6/SvEvTac-derived embryonic stem cells. Resultant mice were crossed with C57BL/6 background FLPe mice to excise the neomycin cassette and backcrossed to C57BL/6 for at least 5-10 generations by the donating laboratory.

Site of ExpressionIn adult mice, Ntrk2 is preferentially expressed in brain tissue, although significant levels of Ntrk2 RNA have also been observed in lung, muscle and ovaries. In situ hybridization analysis of 14 and 18 day old mouse embryos indicates that Ntrk2 transcripts are localized in the central and peripheral nervous systems, including brain, spinal cord, spinal and cranial ganglia, paravertebral trunk of the sympathetic nervous system and various innervation pathways.

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.
